She was born in Sydney, Australia to a Singapore-Chinese mother and Japanese father. She trained in ballet, jazz, and contemporary from a young age, eventually turning to musical theatre and then screen acting.
Kimie Tsukakoshi made her main stage debut in The Dead Devils of Cockle Creek at La Boite Theatre Company in early 2018. She has had many other onstage credits. At the Brisbane Festival, she even performed the titular role in the world premiere of Hanako: Desire and Other Secret Weapons which is by Belloo Creative (the team behind the award-winning play Motherland). Hanako was also nominated at the 2017 Matilda Awards for plenty of awards, one of which was Best Female Actor for Kimie Tsukakoshi.
